Our lab uses a GE Healthcare/Amersham Biosciences Typhoon Trio Imager for high resolution imaging of two dimensional gels. This instrument is capable of imaging with ultra high sensitivity; it is also equipped with multicolor detection. The Typhoon Imager Trio can be used for such imaging applications as: nucleic acid stains, Northern and Southern Blot detection, protein gel stains, and multipurpose fluorescent labels. Multipurpose detection allows the Typhoon to detect multiple fluorescent labels contained within the same gel, which is the basis for Difference In-Gel Electrophoresis (DIGE).
Our facility uses two software programs for gel spot detection and analysis. DeCyder software is a program tailor-made for DIGE experiments. This program is a fully automated analysis software suite for detection, quantitation, positional matching and differential protein abundance analysis. This software is used for the analysis of spots detection within pooled CyDye labeled 2-D gels. For regular 2-D gel spot analysis our lab uses ImageMaster 2D Platinum Software. This program is very similar to the DeCyder software, but used for singly labeled or stained gels. Both programs are powerful tools in differential spot analysis between multiple gels.
