Agilent's HPLC-Chip is the first microfluidic chip-based device that can carry out nanoscale HPLC. The reusable chip, smaller than a credit card, integrates sample enrichment and separation with a spray tip etched directly on the chip. Most of the failures in a nanoflow system are due to leaks and clogs due to fittings and connections. More than half of the connections typically required are eliminated using the chip, therefore dead volumes are reduced and ease-of-use and reliability are increased. Early data suggests that sensitivity and peptide coverage are enhanced on the chip as compared with a traditional nanoflow system. The HPLC-Chip interfaced to an XCT Plus trap is a centerpiece of our analysis of two-dimensional gel spots and is also appropriate for more complex mixtures such as immunoprecipitations.
