ISOLATING THE MITOCHONDRIA

Kits are available from Sigma and Pierce for isolating and enriching mitochondrial proteins from both tissue and cell culture systems. Please follow the links to the protocols. Before using the protocols you will generally:
  1. Pellet cells at 500 x g for 3 min. (Make sure you pre-weigh your tubes so you can determine a cell weight).
  2. Remove supernatant and wash 3-4 times in wash buffer. Remove all supernatant and re-weigh cells to obtain a wet weight.
