Antigen retrieval

 

 

To make:  Sodium Citrate (10mM, pH 9.0) make this up in dH2O.  This solution will keep for several weeks at 4°C

 

Heat hot water bath to 80°C

 

Put coplin jar (or other container that can hold slides) in hot water bath for several minutes before adding slides to solution.

 

Let slides incubate in sodium citrate at 80°C for 5-30 minutes (I usually do 20 minutes)

 

3 x 10 minute washes in PB or PBS (I usually warm my first wash to about 40°C then use room temperature solutions for the last two washes)

 

Continue with immuno protocol

 

 

 

 

 

* If you have problems with tissue coming off of the slides, dip them in water and dry them on a hot plate for a couple minutes prior to doing the antigen retrieval...this may help sections stick to slides.
