PROTOCOL FOR SUBBING SLIDES

1. Load slides into metal racks. Save slide boxes for later use.

2. Place racks into 95% ethanol container in hood. Let sit overnight.

3. Remove racks and place into clean and well rinsed dish pan.

4. Rinse slides in deionized water for 20 minutes

5. Prepare a solution of .5% Gelatin and .05% chromium potassium sulphate

for 500 mls of chrom-alum:

  1. Heat 500 mls of good deionized water to 60° C, using a thermometer to be sure not to exceed 60° C.
  2. Add 2.5 grams of gelatin and allow to dissolve using a stir bar.
  3. Add .25 grams of chromium potassium sulphate and allow to dissolve.
  4. Filter solution, with standard filter paper, into a rectangular staining dish.

6. Fill a rectangular staining dish with good deionized water.

7. Remove each rack of slides using a slide rack holder and dip into the deionized water for 20 seconds, then into the warm gelatin-chrom-alum solution for 20 seconds.

8. Slides should be allowed to dry overnight at not more than 37 C in a dust free location.

9. Place slides back into boxes, label boxes with date and "Subbed Slides", and store in refrigerator.
